In Silico Study of Mangosteen Fruit (Garcinia mangostana L.) as Pancreatic Anticancer Against AKT Kinase

Keganasan pada pankreas diakibatkan karena tidak terkendalinya proliferasi sel yang akan mengarahpada berkurangnya proses apoptosis sel. Oleh itu, salah satu usaha untuk menangani kasuskanker ini adalah dengan menghambat protein antiapoptosis seperti AKT kinase. Inhibisiprotein kinase didasarkan mekanisme penghambatan fosforilasi padasel pankreas. Senyawa golongan xanton dari buah manggis (Garcinia mangostana L.) telah terbuktimemiliki aktivitas antikanker. Pada penelitian senyawa santon diteliti potensinya sebagaiobat anti kanker melalui metode penambatan molekuler dilakukan terhadap reseptorprotein bantuan AutoDockTools (versi 4,2.6 dan 1.5.6), divisualisasikan denganBIOVIA Discovery Studio 2020, lalu analisis profil farmakokinetika serta toksisitas drug-likenessyang mengacu Lipinski’s Rule of Five situs web pre-ADMET. Penambatan molekulermenghasilkan 5 turunan memenuhi kriteria. 1-isomangostin memilikipotensi lebih baik jika dibandingkan ligan pembanding uji lainnyadengan energi ikatan (ΔG) -9,90 kkal/mol konstanta inhibisi 54,9 nM. Hasil prediksi ADMET dari1-isomangostin menunjukkan sifat absorpsi (HIA = 94%, Caco2 39,97), distribusi yangbaik (PPB 79,1%), mampu menembus sawar otak (BBB 0,179), bersifat toksik.
